Why Reconstitution Matters

Most research peptides arrive as a lyophilized (freeze-dried) powder. To use them in research, they must be reconstituted — dissolved in a sterile solution. Proper reconstitution technique directly affects peptide stability and research accuracy.

What You Need

  • Lyophilized peptide vial
  • Bacteriostatic water (BAC water) — contains 0.9% benzyl alcohol as preservative
  • Insulin syringes (29-31 gauge)
  • Alcohol swabs

Step-by-Step Process

Step 1: Gather Materials Ensure everything is clean and the peptide vial has been at room temperature for 10-15 minutes (to reduce thermal shock).

Step 2: Calculate Your Volume The amount of BAC water you add determines the concentration. For example: 5mg peptide + 2mL BAC water = 2.5mg/mL (2500mcg/mL). 5mg peptide + 1mL BAC water = 5mg/mL (5000mcg/mL)

Step 3: Add BAC Water Draw your calculated volume of BAC water into an insulin syringe. Insert the needle into the peptide vial and let the water slide down the inside wall of the vial — never squirt directly onto the powder.

Step 4: Swirl Gently Once the water is added, gently swirl the vial until the powder is completely dissolved. Never shake — this can damage the peptide chains.

Step 5: Store Properly Reconstituted peptides should be stored at 2-8°C (standard refrigerator). Most reconstituted peptides remain stable for 14-28 days.

  1. Squirting water directly onto powder — can damage peptide bonds
  2. Shaking the vial — denatures the protein structure
  3. Room temperature storage — dramatically reduces shelf life
  4. Using sterile water instead of BAC water — no preservative means higher contamination risk for multi-use vials
